Fusarium oxysporum f. sp. cubense (Foc), causes the Panama disease, a major constraint of banana habe been described. Race 4 is also divided into subtropical (ST4) and tropical (TR4) variants. Considering that at least 20 vegetative compatibility groups were identified in Foc and the narrow range of banana cultivars used for race discrimination, it is necessary develop tools to better understand race-cultivar interactions. Transgenic fungi expression reporter genes have been successfully used in plant-pathogen interaction studies and could also be a useful tool for banana- Foc interaction. We have used the Agrobacterium-mediated transformation system to develop strains from four Foc races expressing the reporter genes gfp (green fluorescent protein) or dsred (red fluorescent protein). Two gene constructs carrying either reporter genes driven by the construtive promoter PtoxA were used. Strains carrying gfp of dsred were generated for all the races. Banana-Foc interaction studies using such reporter gene-labelled strains are currently in progress. |
